人心肌肌凝蛋白轻链(CMLC)的分离及临床应用

摘    要:从人心肌提取了肌凝蛋白,并检测了其ATP酶活力的稳定性。用SDS聚丙烯酰胺凝胶电泳(SDS-PAGE)法纯化并鉴定了人心肌肌凝蛋白轻链(CMLC),结果表明CMLCI的分子量为27000;CMLCⅡ的分子量为20 000。它们的紫外吸收光谱显示A260>A280,表明其高含苯丙氨酸。制备了兔抗人CMLC抗血清,免疫双扩散的结果表明,纯化后的CMLC与肌凝蛋白有相同的抗原性。用阳性的兔抗人CMLC抗血清为对照作ELISA实验,不仅从免疫学角度进一步证实了人心肌CMLC的特性且为其在临床诊断应用提供了可靠的方法。

Abstract:Cardiac myosin was extracted from normal human cardiac muscle.The specific activity of ATPase of cardiac myosin was assayed for demonstrating it.As stability.The CMLC was purified and identified by SDS-PAGE The results showed that the molecular weight of CMLC *********I is 27kD and the molecular weight of CMLC ***********II is 20kD- The ultraviolet spectra of CMLC showed A260>A280.This result suggested that CMLC contains many phenylalanines.We have prepared the rabbit antiserum against human CMLC The immune-double diffusion pattern showed that CMLC possesses same antigenicity with myosin.ELISA was carried out by using the positive rabbit antiserum against human CMLC as control.All these results demonstrated not only the immanological characterization of human CMLC but also offered a reliable method for clinical examination of the myocardialinfarction.
