二氢玉米素核苷组细胞分裂素的放射免疫测定法

本文报道二氢玉米素核苷（ＤＨＺＲ）组细胞分裂素放射免疫测定法（ＲＩＡ）的研制结果。以（３Ｈ）二氢玉米素作示踪剂，测得免抗ＤＨＺＲ抗血清的效价为１：１３７０（Ｂ％＝３０％）。该抗血清主要与本组细胞分裂素发生交叉反应。回收率为９６．９％，灵敏度为１２ｆｍｏｌ ＤＨＺＲ／管，检测线性范围为０．１￣１００ｐｍｏｌＤＨＺＲ／管。批内误差ＣＶ＝４．３，批间误差ＣＶ＝２．０％。对基于同一免抗血清的ＤＨＺＲ组细胞

Radioimmunoassay for Dihydrozeatinriboside-group Cytokinins

Radioimmunoassay for dihydrozeatinriboside-group cytokinins(mainly including DHZ,DHZR) has been developed.Dihydrozeatinriboside(DHZR) was selected as hapten and coupled to bovine serum albumin(BSA) via NalO4 oxidation,then synthesized immunogen DHZR-N,-BSA.Antisera against this immunogen was raised in New zealand white rabbits.Its titer,determined by use of(3H)DHZ as tracer,reaches 1:1370(B% =30%).The antisera cross-react mainly with DHZR-group cytokinins.Recovery(3H)DHZ as interstandard) is 96.9%.Sentivity is 12 femtomol DHZR/tube.Linear detection range is 0.1 100 picomol DHZR/tube.Coefficient of variation within batch is 4.3% and that between batches is 2.0%.After compared a variety of parameters(including antisera cossreactivity,assay parameters,recovery,validity,etc.) with those of DHZRs ELISA based the same antisera(Chen Y-F,et al,J Nanjing Agric Univ,15(2):122-6,1992),it is concluded that these parameters of DHZRs RIA and DHZRs ELISA are basically agreeable,and that DHZRs contents in corn grains determined with the two immunoassay are comparable.
