家兔胚胎冷冻保存——AgI植冰法的应用

摘    要:以日本大耳白兔为供胚动物,采用FSH-p加hCG超排。于交配后62 ̄65小时手术或屠宰,冲洗子宫角及输卵管采得正常的桑椹胚402枚。用两种植冰方法和两种装管顺序将兔胚置于塑料细管进行慢速冷冻。结果AgI植冰法与传统的植冰法具有同样的植冰效果,其快速解冻后胚胎的可移植率分别为85%和80%;培养48小时后发育至扩张囊胚的比例分别为90%和86%,均显著高于未植冰的两种装管方式(P〈0.01)。AgI

Abstract:The aim of present study was to develop an automatic and reliable seeding method byusing silver iodide(AgI) as the ice nucleator. The experiment was carried out on 402 morulastage rabbit embryos. Two methods of seeding and two methods of loading embryos intoplastic straws (total four groups) were used. The effectiveness of AgI was evaluated by invitro survival After thawing, the rate of trasferable embryos in four groups were 85, 10,80and 15% respectiviy, and the rate of embryos developed into expanded blastocysts were 90,13,86 and 18% respectiviy. The transferable and development rate of AgI seeding method(group A) was same as the conventional seeding method (group C), and higher than othergroups (B and D) without seeding. The seeding methed by AgI can be useful in the cryopreservation of rabbit embryos.
