以合成多肽为抗原的戊肝抗体检测试剂的研制

用戊肝病毒（ＨＥＶ）基因组编码氨基酸序列１－９０１－９１４／２－５１５－５３０、３－９１－１２３、２－６１３－６５４相应的三段合成多肽为抗原、研制出一种检测抗－ＨＥＶＩｇＧ的ＥＬＩＳＡ试剂。以该试剂检测中国、缅甸、印度和前苏联肠道传播非乙型肝炎（ＥＴ－ＮＡＮＢＨ）病人血清１０５份，仅３份中国病人血清阴性，阳性率为９７．１％；检查实验感染ＨＥＶＬ赤猩猩血清，感染前阴性，感染后阳性；检查正常人血清９９

Establishment of a Sensitive ELISA for Detecting Antibodies against hepatitis E Virus Based on Synthetic Peptides

sensitive ELISA for detection of antibodies against hepatitis E virus(anti-HEV)was established,based on three synthetic peptides from open readin franmes ORF1,ORF2 and ORF3 of the HEVgenome.sera were tested for anti-HEV with our ELISA,One hundred and tWo(97.1%)out of 105sera from patients with enterically transmitted non-A non-B hepatitis E in Chil1a,Burma,India and former USSR were tested positive.A total of 99 sera from normal individuals in Beijing were tested negative.sera from experimentally infected chimpanzee with HEV wete positive,but pre-infection serumfrom the anllnal was negative,Of 4 6 sera from potients with hepatitis A,B and C,only one from pa-tient with hepatitis C was positive.Between our(AMS)and Diagnostic Biotechnology(DB)anti-HEVELBAs,the differences of sensitivity and specificty were not significant,the rate of consistericy was81%.All epitopes involved in the two ELlSAs may not be identical,and it might be responsible for thedifference.
