胡黄连药材的指纹图谱研究

摘    要:利用HPLC—DAD—MS法,研究胡黄连的高效液相色谱指纹图谱,为科学评价及有效控制其质量提供可靠方法。采用C18色谱柱,甲醇-水(含0.1%的甲酸)梯度洗脱;流速为1.0mL/min,检测波长290nm;柱温为室温。对不同产地的胡黄连药材进行指纹图谱分析,通过HPLC—MS技术指认指纹图谱共有模式中共有峰的归属。比较5种产地胡黄连药材指纹图谱确定了16个共有峰,并指认了含量超过50%的胡黄连苷Ⅱ和苷Ⅰ。发现产地不同,其指纹图谱有差别。试验方法和结果具有较好的准确性和稳定性,可以为胡黄连药物研究提供参考资料,对胡黄连药材的质量评价具有重要的参考价值。

Abstract:The fingerprint of Picrorhiza scrophulariiflora Pennell by HPLC-DAD-MS was used to establish a sensitive and specific HPLC method for quality control. The experimental conditions were as follows: C18 chromatographic column, mobile phase of MeOH-H20(both include 0. 1% methanoic acid), flow rate at 1.0 mL/min, gradient elution, detection wavelength at 290 nm and room temperature. The fingerprints of Picrorhiza scrophulariiflora Pennell from different places were analyzed and the same peaks were identified by HPLC/MS. Sixteen common peaks were detected among 5 different samples. At the same time, the picroside Ⅰ and Ⅱ were identified, whose total contents in samples is above 50%. The analytical method is simple, effective and feasible and can be used to inspect the quality of Picrorhiza scrophulariiflora Pennell.
