纳帕海高原湿地噬藻体psbA遗传多样性

摘    要:噬藻体在水生系统中分布广泛,数量丰富,具有丰富的生物多样性。为揭示纳帕海高原湿地噬藻体psbA遗传多样性,并分析其系统进化地位。本研究于纳帕海高原湿地采集土壤样品,以特异性引物对其光合基因psbA进行扩增,得到不同的psbA基因序列51条,并在淡水、海水、稻田等不同环境下进行PCoA分析和系统发育分析。通过研究得知,在纳帕海高原湿地环境下的psbA基因和在稻田环境中的psbA相接近,而与海洋环境相距较远,并且大多与蓝藻细菌相聚簇,而且psbA基因中的NT-4和NT-7与噬藻体具有较高相似性,和其他地区比较较远。推测NT-4和NT-7是纳帕海高原湿地特有的噬藻体类群。

Abstract:Cyanophage are widely d istributed in aquatic systems, which are abundant in quantity and rich in biodiversity. In order to reveal the genetic diversity and analyze the phylogenetic position of cyanophage psbA in the Napahai plateau wetland, the photosynthetic gene psbA was amplified by using specific primers with the soil samples gathered from Napahai plateau wetland, and then 51 different psbA gene sequences were obtained. The phylogenetic analysis and PCoA analysis were studied with other habits such as marine, freshwater and paddy field. It showed that the psbA gene in the Napahai plateau wetland was close to that in paddy field, as well as was far away from that in marine, of which most were clustered with cyanobacteria. Besides, NT-4 and NT-7 in psbA gene had highly similarity with cyanophages, while they were far away from other areas. It was speculated that NT-4 and NT-7 were unique cyanophages in Napahai plateau wetland.
