卵巢癌DKK-1的表达与肿瘤转移的相关性研究

目的：探讨DKK-1在卵巢上皮癌和淋巴结转移癌中表达及其与肿瘤侵袭的关系。方法：应用免疫组化SABC法，对80例卵巢上皮肿瘤手术标本进行DKK-1蛋白表达的测定。结果：DKK-1在卵巢上皮癌中阳性表达率为70．9％，其明显高于良性肿瘤及正常组织（P〈0．01），有淋巴结转移的肿瘤的阳性表达率为46．7％，无淋巴结转移的肿瘤阳性表达率为80．0％，DKK．1低表达与卵巢癌侵袭和淋巴结转移关系密切（P〈0．05）。不同细胞分化等级与临床分期间DKK-1表达无明显差异（P〉0．05）。结论：DKK—1蛋白的表达下降可能是上皮性卵巢癌发生转移的一个早期事件，有望成为预测卵巢癌发生转移及判断患者预后的参考指标。

Correlation of the DKK-1 and the metastasis of human epithelial ovarian carcinomas

Objective： To investigate the expression of DKK-1 protein in human ovarian armor and lymph node with metastasis and its relationship with the invasion. Methods： SABC immunohistochemical method was used to detect the DKK-1 protein expression in 80 cases with human ovarian tumor after surgical treatment. （55 cases ovarian cancer, 25cases benign ovarian tumor）. Results： In 55 ovarian cancer tissues, the expression rate of DKK-1 were 70.9%, the expression rate of DKK-1 in benign ovarian tumors were 16.0%. The two groups had obvious difference （P〈0.01）. A significant correlation was found a low DKK-1 expression rate in the invasion and the lymph node metastasis of ovarian cancer （P〈0.05）. The expression of DKK-1 protein had no statistical correlation with cell differentiation and clinical stages （P〉0.05）. Conclusion： Expression of significantly increase in ovarian cancer and they are related to ovarian invasion and. The decrease of DKK-1 protein expression might frequently occur in early stage of metastatic tumors and the expression of DKK-1 protein might be used as a prognostic marker which indicates the potential of ovarian epithelial cancer.
