包囊游仆虫包囊形成和解脱过程中大、小核的研究

摘    要:包囊游仆虫形成包囊时大核除经历形态大小的变化外,大核DNA含量也低于正常大核水平;细胞脱包囊前,在大核一端或两端发生染色质粗浓集,是大核DNA复制的结果;染色质粒在整个大核内浓集时,大核DNA含量已达到正常大核水平,此时DNA复制结束,细胞脱包囊。小核在游仆虫形成包囊和脱包囊过程中,其形态大小、DNA含量等无明显变化。

Abstract:By means of optical microscopy, it was observed that during encystment and excystment of Euplotes encysticus the macronucleus (Ma) of a resting cyst was circular in shape and the chromatin granules were few and scatered. Before excystment, there first appeared dense chromatin granules at one or both ends of the Ma and then the whole Ma was filled evenly throughout with dense chromatin granules. Sometimes, some of the chromatin granules were further condensed into chromatin spheres. After excystment, the Ma and its chromatin granules returned to its normal morphology.From feulgen reaction specimen, the three-dimensional absorbing graph and quantitative data of microphotometry results were obtained. It was clearly shown that the Ma had undergone changes in appearance and size during encystment and excystment. Besides there was a lower DNA content in the Ma of a resting cyst than the Ma of a normal cell. The appearance of dense chromatin granules at one end or at both ends of the Ma was related to the replication of Ma DNA. By the time when the Ma was full of dense chromatin granules the DNA content had reached the level of a normal Ma; and when DNA replication in the Ma was completed, excystment occured.On the basis of the above observations, the author presumes that these changes in DNA content of Ma are obviously related to the encystment and excystment processes. Because no obvious changes were observed in the micronucleus (Mi), it seems that the Mi is not involved in the encystment and excystment process.
