泽蛙雌核生殖单倍体产生过程的细胞学研究

摘    要:丁汉波等(1965)在福建无尾两栖类的杂交实验中发现,泽蛙(Rana limnochris Boie)♀×黑眶蟾蜍(Bufo melanostictus Schneider)♂可以产生泽蛙雌核单倍体,但是这种组合的“杂交”如何产生单倍体,在细胞学方面未深入研究。为了进一步了解其发育机理,本文主要从细胞学的角度探讨黑眶蟾蜍精子诱发泽蛙卵进行雌核生殖的过程。

Abstract:We have recently made a cytological study on the formation of the gynogenetic haploid which was produced by a cross between Rana limnocharis (female) and Bufo melanostictus (male). The results are presented as follows:1. The toad sperm in high concentration (2 testes/1 ml 10% Ringer's sol.) was found to be able to raise the rate of the gynogenesis in the combination of the cross.2. A frog egg penetrated by a single toad spermatozoon was observed to excrete a second polocyte, and the egg nucleus was changed into female pronudeus and moved towards the central area of the animal pole. The sperm nucleus was neither changed into a male pro-nucleus nor merged with the female pronudeus but was expelled from the egg through the first cleavage furrow. The proximal centriole (oblique centriole) of the spermatozoon played an important role in the gynogenesis, as it interacted with the cytoplasm of the egg to form a spermaster, and subsequently, the spermaster became divided (or duplicated) into two, forming an amphiaster to meet the needs of the gynogenesis.3. A new staining method (gentian violet-light green double colouring method) for the study of the cytological process of fertilization, which has brought about a good effect, would also be described.
