摘    要:以花楷械、青楷械和小楷械的嫩茎段为外植体,应用均匀设计法筛选最适合于嫩茎腋芽生长伸长同时生根的培养基.结果表明,最适合花楷械的培养基为:改良MS+IAA0.10mg·L^-1+GAa1.10mg·L^-1,再生率为98%;最适合青楷械的培养基为:改良MS+IAA0.40mg·L^-1+IBA0.10mg·L^-1+NAA0.07mg·L^-1+GA31.10mg·L^-1,再生率为96%;最适合小楷械的培养基为:改良MS+IBA0.10mg·L^-1+GA31.10mg·L^-1,再生率为94%.以再生植株的茎节为材料进行快繁的结果表明,在30-35d的一个培养周期内,增殖倍数达65以上.

Uniform Design Methods for High Efficient Micropropagation System Optimization of Three Plants of Acer

Abstract:Young stems of Acer ukurunduense Trautv. et Mey., Acer tegmentosum Maxim., and Acer komarovii Pojark. were used as explants and suitable medium compositions for axillary bud growing and rooting were screened through Uniform Design experiments. The results showed that MS (modified)+IAA0.10 mg·L^-1+GA31.10 mg·L^-1was most suitable for Acer ukurunduense with a regeneration rate of 98%; MS (modified) + IAA0.40 mg·L^-1+IBA0.10 mg·L^-1+NAA0.07 mg·L^-1+GA31.10 mg·L^-1was most suitable for Acer tegmentosum with a regeneration rate of 96%; MS (modified)+IBA0.10 mg·L^-1+GA31.10 mg·L^-1was most suitable for Acer kornarovii with a regeneration rate of 94%. Stems each with one node were cut from regenerated shoots and cultured for propagation, and a 65-fold proliferation rate was achieved within 30-35 days.
