翘鳞香菇信息素受体编码基因的克隆及其进化分析

摘    要:翘鳞香菇是一种新近开发的具有很大市场潜力的食用茵新品种,但是对翘鳞香菇的分类地位还存在有一定的不确定性。本研究结合简并PCR和染色体步行技术克隆获得了翘鳞香菇B交配型位点中的信息素受体编码基因,然后对翘鳞香菇以及其它15种担子菌的信息素受体的氨基酸序列进行了系统分类学分析,结果显示,翘鳞香菇与香菇具有很近的亲缘关系。这个结果与运用ITS序列对包括翘鳞香菇在内的16种担子菌进行的系统分类学分析结果基本一致。本研究从系统分类学角度为判断翘鳞香菇的分类地位提供了丰富的实验证据。

Abstract:Lentinus squarrosulus with big market potential is one kind of recently developed fungus strains. However, the classification of Lentinus squarrosulus is uncertain. The degenerate PCR and the chromosome walking technique were used to reveal the molecular genetics structure of pheromone receptor gene LSrcb1-B1 in B mating type gene in strain No. 7-4 of Lentinus squarrosulus . After bioinformatic analysis, phylogenetie analysis on amino acids of pheromone receptor gene LSrcb1-B1 as well as other 15 kinds of basidiomyeetes was done by using the NJ method in PHYLIP software package. The result showed that the Lentinus squarrosulus shared the same cluster with the Leneinula edodes. This result was also supported by ITS analysis of 16 kinds of basidiomycetes. This research provided a rich experiment evidence in Lentinus squarrosulus classification and phylogenetie analysis.
