Abstract:The off20 in the ramoplanin biosynthesis gene cluster shows high homology to the known halogenases. An inframe knock-out plasmid pSM-3 was transformed into Actinoplanes sp. ATCC 33076 to delete the off20 gene fragment encoding 64 amino acids. A representative off20 mutant was designated SIPI-A. 2001 dorf20 (△orf20). Absence of ramoplanin and its analogs production from the resulting exconjugants suggested that the protein coded by off20 was a key enzyme in the ramoplanin biosynthesis pathway, and it might be have other functions except halogenation.
