摘    要:以Penicillium sp.B01为出发菌株,经吖黄素或DES(硫酸二乙酯)分别与^60Co-γ射线对其孢子悬液进行复合诱变。经过初筛和复筛,在30μg/mL吖黄素诱变时间2h,剂量率为4.11Gy/min的^60Co-γ射线辐射使累计剂量为20.55Gy复合诱变的条件下,筛选出一株菊粉酶活比出发菌株高32%的突变菌株B01-A13-Co31。经同工酶电泳验证,变异株与出发菌株相比酶带有所变化。将此菌种连续传代6次进行产酶性能的稳定性测定,表明此菌株具有良好的遗传稳定性。

Abstract:The spore suspension of original strain Penicillium sp. B01 was mutagenized by DES-^60Co-γ-rays and acriflavine-^60Co-γ-rays respectively for screening highly productive inulinase mutant strain. After primary and second screening, the inulinase activities of the productive mutant strain increased by 32 % as compared to that of the original strain. The highly productive inuliase mutant strain was screened and obtained from a large number of mutant samples under the conditions of 30 g/mL acriflavine for treating for 2 hours,^60Co-γradiation at the irradiation dose of 20.55 Gy with the dose rate of 4.11 Gy/min. By analysis of the isozyme electrophoresis, the bands of mutant strain had some changes as compared to that of the original strain, indicating that they were homologous but heterologous. It was demonstrated that the mutant strain had a stable transmissibility for 6 generations with high productivity.
