Abstract:The breeding of high brefeldin A-producing strains and optimization of fermentation conditions were investigated. Eupenicillum sp. E-0506 used as an original strain for mutagenetic breeding by LIV, NTG and UV + NTG, respectively. The fermentation conditions were optimized by single factor test and orthogonal test. The spore sus- pension of E-0506 was treated by UV irradiation for 60 seconds, followed with 2.0 mg/mL NTG for 30 minutes under magnetic stirring. A mutant Eupenicillum sp. E-UN88 producing brefeldin A (BFA) with high yield and well hereditary stability was obtained. Its optimum fermentation parameters were as follows: medium volume 100 mL/500 mL, inoculum size 5 %, cultivation time of seed 43 h and fermented at 26 ~2, 250 r/min, for 120 h. The BFA yield of Eupenicillum sp. E-UN88 was enhanced more than 10.36 times higher than that of the original strain. The results laid a foundation of the brefeldin A production of industrial fermentation.
