培养方式对盐藻生长和积累β-胡萝卜素的影响

摘    要:盐藻细胞生长和积累β-胡萝卜素的最佳条件存在差异,通过正交实验获得盐藻生长最适浓度C、N、P的分别为15、2.0、0.2mmol/L,累积β-胡萝卜素最适浓度分别为15、1.0,0.1mmol/L。比较了一次添加型、分次添加型、不完全更换型和完全更换型4种培养方法对生长和累积β-胡萝卜素的影响,发现完全更换型培养方式有利于β-胡萝卜素的积累。中途补给10mmol/L NaHCO3也有利于藻细胞积累β-胡萝卜素。在实验最佳条件下藻液中的β-胡萝卜素含量是对照的1.43倍。可采用先快速培养盐藻细胞、后更换培养基、添加NaHCO3分段培养方式以促进细胞大量合成β-胡萝卜素。

Abstract:The optimum conditions for the growth and β-carotene accumulation of DunalieUa salina cell were different. By the orthogonal experiment the optimum concentrations of carbon source, nitrogen source, phosphate source for cell growth were 15, 2.0, 0.2 mmol/L , respectively, while the optimum concentrations for the β-carotene accumulation were 15, 1.0, 0.1 mmol/L respectively. Four cultivation methods of traditional batch method, fractional addition, complete replacement and incomplete replacement were compared with the cell number and β-carotene accumulation as parameters. The data showed that complete replacement method was beneficial to the β-carotene accumulation. It was found that the adding of NaHCO3 in the middle way of cultivation increased the accumulation of β-carotene. Under the optimum conditions the β-carotene content of D. salina was 1.43 times of the control sample. So for the production of β- carotene the steps were to obtain the fast cell growth first, then to design suitable replacement of the medium and NaHCO3-adding.
