| 摘 要： | CDF家族蛋白在金属离子的动态平衡和耐受过程中起重要作用,拟南芥基因组序列分析表明,拟南芥中CDF家族至少由12个成员构成.本研究通过功能互补的方法研究其中的一个成员AtMTP6的功能.应用RT-PCR克隆得到其cDNA,克隆至pMD18-T载体并测序,结果显示与AtMTP6的ORF序列一致.然后亚克隆至表达载体pTrc99A并转化至大肠杆菌突变株KAM3.IPTG诱导表达后进行功能互补分析,结果表明AtMTP6能转运锌离子进入KAM3,使细菌对高浓度的Zn2 更加敏感.

| Abstract: | Cation diffusion facilitator(CDF) proteins play an essential role in metal homeostasis and tolerance.Analysis of the Arabidopsis genome identified no less than 12 genes that encode proteins belonging to the CDF family.One of the member AtMTP6 was studied using functional complementation analysis.The AtMTP6 cDNA was amplified by RT-PCR,cloned to pMD18-T vector and sequenced.It showed the same as the AtMTP6' s ORF.Then cDNA was subcloned to pTrc99A and transformed to the E.coli mutant KAM3.Complementation assay showed that AtMTP6 transport Zn2 into the cells.It made the cells more sensitive to the excessive Zn2 . |
