平菇产植酸酶菌株的筛选及植酸酶基因区域的克隆

摘    要:从平菇(Pleurotus ostreatus)8个菌株中筛选出3株高产植酸酶菌株,并根据GenBank中植酸酶基因的保守区设计并合成一对特异性引物,以平菇菌丝的总DNA为模板,通过PCR扩增,获得了一条长约920 bp的片段.DNA序列测定结果表明,该片段长度为919 bp.采用blast进行序列比对,结果表明:该片段与曾报道的源于Trametes pubescens的植酸酶phyA(GenBank Accession:AJ310700)基因相比较,其DNA序列同源性为93%.该片段含有3个内含子,含有植酸酶基因的活性位点保守序列(Active-site sequence)RHGARYPT.

Abstract:Three Pleurotus ostreatuses that producing largely phytase were sc re ened from the eight kinds of Pleurotus ostreatuses.The phytase gene region was cloned by PCR amplification from Pleurotus ostreatus chromosomal DNA with a pair of specific primers designed according to the conserved sequence of phytase gen e in GenBank.DNA sequencing results showed that the fragment was 919 bp.Sequen ce analysis with blast search indicated that the DNA sequence had 93%homology i n nucleotides,comparing with the reported phytase phyA(Genbank Accession: AJ310 700) from Trametes pubescens.In addition,it contained three introns and the ac tive-site conserved sequence RHGARYPT.
