摘    要:程序性细胞死亡分子5(programmed cell death 5,PDCD5)在人体各种组织中广泛存在,可以通过多种凋亡通路促进细胞凋亡,其在大部分肿瘤中低表达,对肿瘤化疗具有增敏效应。近年发现,除了在细胞凋亡过程中发挥作用外,PDCD5在多种疾病的病理进程中发挥重要作用,如:肿瘤、自身免疫性疾病、炎症性疾病、脑缺血再灌注损伤及动脉粥样硬化等。PDCD5蛋白结构简单,无二硫键,其重组蛋白药物开发较便利,具较高的药物基因组学应用价值。本文主要介绍近20年来PDCD5的研究进展及其在肿瘤、免疫系统疾病、心血管系统疾病中可能的应用、存在的不足、以及在其他方面的应用前景。

Abstract:Programmed Cell Death 5(PDCD5)is ubiquitously expressed in various human tissues,promoting cell apoptosis through multiple apoptotic pathways;PDCD5 has low expression level in most kinds of tumors,and it can increase the sensitization of tumors in response to chemotherapy.Increasing evidence showed that,in addition to participating in cell apoptosis,PDCD5 plays important roles in a variety of diseases,such as cancers,immune system diseases,inflammatory diseases,ischemia reperfusion injuries,and atherosclerosis.The structure of PDCD5 protein is quite simple without disulfide bond,therefore,it is very convenient to develop and produce the recombinant protein for therapeutic purpose in the future.This review mainly focuses on the research advance of PDCD5 in the recent two decades,the potential application and defects of PDCD5 as therapeutic strategies for cancers,immune system diseases and cardiovascular diseases,as well as perspectives of the value of PDCD5 in other directions.
