稻米赖氨酸快速测定条件的探讨

摘    要:以氨基酸自动分析仪测定稻米赖氨酸含量为参照标准。在“茚三酮测定赖氨酸含量”(A法)的基础上,对该法中大米蛋白质的提取温度和时间、色温度和时间及茚三酮用量等条件进行了探讨,提出了稻米赖氨酸含量快速测(B法)。提取条件为90℃5分钟,显色条件为90℃20分钟;茚三酮试剂用1 ml。用B法对10个大米样品进行测定,赖氨酸含量与氨基酸分析仪测得的赖含量基本上一致,无显著差异(P>0.05)。

Abstract:"Raoid lysiae determination with ninhydrin" method A has been irtvestigated, using rice lysine content assayed by Automatic Amino Acid. Analyser as a control. The conditions for method A such as extraction temperature and time of rice proteins, color development temperature and time, and dosage of ninhydrin have been studied. Based on these another rapidrice lysine determination method(method B) was pr oposed, For the method B, the extraction conditions are 90℃ for 5 min; color development conditions are 90℃ for 20 min; the dosage of the ninhydrin is 1 ml. 10 rice samples has been assayed using the method B and the lysine content is similar to those determined by Automatic Amino Acid Analyser, no significant difference existed (P>0.005).
