非小细胞肺癌中Livin和JNK1的表达及相关性研究

摘    要:目的探讨在非小细胞肺癌(non small cell lung cancer,NSCLC)中的Livin、JNKI蛋白的表达及相关性。方法采用免疫组织化学(二步法)检测187例NSCLC肺癌组织Livin、JNK1包括总JN1蛋白(t-JNK1),磷酸化JNK1(p-JNK1)]蛋白的表达情况,采用Spearman等级相关分析探讨其相关性。结果Livin主要在胞浆中定位表达,在肺癌中的表达阳性率明显高于肺良性病变对照组;在鳞癌中表达阳性率明显高于腺癌和其他癌类(P〈0.05)。其在肺癌表达的积分明显高于良性对照组(P〈0.05),其在鳞癌中表达的积分明显高于腺癌及其他癌类组织。t-JNK1、p-JNK1在肺癌细胞上及在假复层纤毛柱状上皮均可见表达,主要在胞浆中定位表达,t—JNKl未见核内定位表达,而p-JNKl偶见核内定位表达。t—JNK1在肺癌组表达阳性率明显低于对照组(P〈0.05),t-JNKl在肺癌中表达的积分低于对照组;而p-JNK1在肺癌组表达阳性率明显高于对照组(P〈0.05)。p-JNK1在肺癌中表达的积分高于对照组,但两者无统计学意义(P〉0.05)。Livin与t-JNK1呈现显著的负相关,Livin与p-JNK1呈现显著的正相关。结论Livin在肺癌中高表达,其在鳞癌中表达高于腺癌和其他类型癌。t-JNK1在肺癌中低表达,而p-JNK1在肺癌中高表达。Livin与p-JNK1表达正相关,推测Livin可促JNK1蛋白磷酸化。

Relationship between Livin and JNK1 Expression in NSCLC

Abstract:Objective Livin, a member of inhibitors of apoptosis protein, is highly expressed in tumor tissues and contribute to tumorigenic development. It is argued that Livin resists apoptosis by JNK1 signaling pathway. This study was aimed to investigate the expression of Livin in non small cell lung cancer (NSCLC) tissues and its relationship to JNK1 expression. Methods Expression of Livin and JNK1, including total JNK1 ( t-JNK1 ) and phosphorylation JNK1 protein ( p-JNK1 ), was detected in 187 NSCLC samples, as well as in 40 lung benign tissues, by immunohistochemical assay. The data were analyzed by Spearman' s rank correlation. Results Livin was mainly expressed in endochylema, rarely in cellular nucleus, while higher expression of Livin was detected in NSCLC tissues than in benign lung tissues ( positive rate : 65.78 % versus 0. 00 % ; immunohisto- chemistry scores: (6.369 ±4. 163) versus (0.475±0.816), P 〈0.05) . Moreover, Livin showed higher expression in Squamocellular tissues than in adenoearcinoma and other tissues (posi- tive rate : 74. 07 % versus 61.96 % versus 42.86 % , immunohistochemical scores : (7. 272 ± 4. 102) versus (6. 000 ±4. 124) versus (3. 571 ±3. 298), P 〈0. 05) . On the other hand, most t- JNK1 and p-JNK1 proteins were detected in the endochylema of both lung cancer tissues and bron- ehus pseudostratified eiliated cohmmar epithelium. In three lung cancer tissues p-JNK1 was detected in the nucleus, t-JNK1 showed no nuclear location and was detected in NSCLC tissues, higher than in lung benign tissues (positive rate: 46. 5 % versus 70 % , immunohistochemical scores: (4. 840 ±4. 264) versus (6. 275± 4. 344) , P 〈 0. 05). Higher expression of p-JNK1 was shown in NSCI,C tissues than in lung benign tissues (positive rate: 58.28 % versus 40 % , immunohisto- chemical scores: (5. 722±4. 177) versus (4. 350 ±4. 306) , P 〉0.05). Expression of Livin dis- played positive correlation with expression of p-JNKl,
