摘    要:钠氢交换子调节因子1(Na+/H+ exchanger regulating factor1,NHERF1)是由358个氨基酸组成的多功能连接蛋白。NHERF1通过其PDZ-1、PDZ-2和EB结构域与多种蛋白质结合,对PDGF、P13K/Akt等信号转导通路起到调控作用。目前研究发现,NHERF1在多种肿瘤中存在异常表达和定位,并且通过与多种抑癌蛋白的相互作用参与肿瘤的发生。文章综述了NHERF1结构、定位、以及在肿瘤(如乳腺癌和肝癌)中的作用机制。

Role of NHERF1/EBP50 in Cancer Occurrence and Development

Abstract:Na+/H+ exchanger regulating factor 1 (NHERFI) is a muhifunctional connectin which is composed of 358 amino acids. It associates with various proteins via its PDZ-1, PDZ-2 and EB domains and acts as a regulator of PDGF and P13K/Akt signal transduction pathway. Recent studies have suggested that NHERF1 is expressed and located abnormally in different cancers. Furthermore, NHERF1 has been found to interact with some tumor suppressor proteins, thus being involved in tumorigenesis. This review introduces the structure and cellular location of NHERF 1, and its role in development of cancer, such as breast cancer and liver cancer.
