摘    要:评估土拉弗朗西斯菌LVS在感染鼠巨噬细胞早期与细胞膜的相互作用。用表达GFP的土拉弗朗西斯菌LVS感染鼠巨噬细胞1774A1。结合单抗的小窝蛋白-1或转铁蛋白受体-1分别用键合了Alexa594的羊抗鼠二抗显色。土拉弗朗西斯菌疫苗株LVS可以诱导宿主细胞膜伸出伪足,将细菌吸收进入巨噬细胞。分布在细胞膜上的小窝蛋白-1和转铁蛋白受体-1参与巨噬细胞对弗朗西斯菌的摄入。这些发现说明,弗朗西斯菌进入巨噬细胞需要细胞膜微结构域和小窝蛋白;在感染早期转铁蛋白受体-1参与了细菌的摄入,这可能与弗朗西斯菌获取铁以利在胞内生存有关。

Abstract:In order to evaluate the interaction between Francisella tularensis LVS and host cell membrane during its invasion into mouse macrophages, GFP expressing F. tularensis LVS was used to infect murine maerophage J774A. 1. Caveolin-1 or transferrin receptor-1 combined with monoelonal antibody was respectively stained with goat-anti-mouse lgG bonding with Alexa 594. The results showed that F. tularensis live vaccine strains could induce host cell membrane to extend parapodia to absorb the cell into maerophages. Caveolin-1 and transferrin receptor-1 distributed on cell membrane involved the uptake of the bacteria. These findings showed that the entry of F. tularensis into maerophages needed cell membrane' s microdomains and caveolin-1, and during the early infection transferrin receptor-1 participate the uptake the bacteria, this may have something to do with the bacteria to obtain iron conducive to the bacterial survival inside the host cells.
