桦褐孔菌提取物对胃癌MGC-803细胞株的抗增殖与诱导凋亡作用

摘    要:探讨了桦褐孔菌提取物对胃癌MGC - 80 3细胞株的抗增殖、诱导凋亡作用及对凋亡相关基因表达的影响。MTT比色法结果显示 ,桦褐孔菌提取物在 0 .5~ 16 0 μg/mL范围内 (其IC50 为 4 μg/mL)对胃癌MGC- 80 3细胞株均有抑制作用 ,并表现出浓度依赖性关系 ;凋亡形态学观察结果 ,药物浓度 2 μg/mL ,作用时间 12~ 2 4h后 ,细胞核染色质固缩并凝结成块 ,聚集在核膜周边 ,凋亡小体形成 ;TUNEL法检测结果 ,不同浓度药物均诱导胃癌MGC - 80 3细胞株凋亡 ,细胞凋亡率随药物浓度增加而上升 ,显示明显的量效关系。Ki- 6 7抗原检测结果 ,不同浓度药物均抑制胃癌MGC - 80 3细胞株增殖 ,表现出浓度、时间依赖性关系 ;2μg/mL桦褐孔菌提取物作用胃癌MGC - 80 3细胞株 4 8h之后 ,明显下调Bcl - 2基因蛋白表达。因此 ,通过此项研究可得出 ,桦褐孔菌提取物对胃癌MGC - 80 3细胞株有抗增殖作用和诱导凋亡作用 ,其凋亡的分子生物学机制可能与下调凋亡抑制基因Bcl 2表达有一定关系。

Abstract:The effects of the extractions from Inonotus obiquus (Fr.) Pilat on anti proliferation, induced apoptosis and gene expressions for MGC 803 cells of gastric cancer were investigated. The extracts from the fungus had the inhibiting action on MGC 803 cells of gastric cancer line in the concentration of 0.5~16 μg/mL (IC 50 is 4 μg/mL) by means of MTT assay, and exhibited a strong dependence on concentration. Morphologic changes of cell apoptosis were observed in the concentration of the extract in 2 μg/mL in 12~24 h; TUNEL showed that the extracts in different concentrations had significant quantity dependent effects on apoptosis induced MGC 803 cells of gastric cancer cell line, and the effects would increase as the concentrations increased; Ki 67 anti proliferation test indicated that the extracts of various concentrations all had concentration and time dependent effects on apoptosis induced on MGC 803 cells of gastric cancer line; The expression of oncogene Bcl 2 decreased after the administration of the extracts at a concentration of 2 μg/mL for 48 h. The extractions of Inonotus obiquus (Fr.)Pilat have been shown to have effects on anti proliferation and apoptosis inducement, and the molecular biological mechanism is related to suppressing the expression of oncogene Bcl 2.
