摘    要:以悬钩子皮下盘菌等总基因组DNA为皮下盘菌属(Hypodermade Not.)及其近似类群RAPD体系优化的模板,对模板DNA浓度、dNTPs浓度、Mg2+浓度、引物浓度、TaqDNA聚合酶浓度等反应体系以及退火温度和时间、延伸时间、循环次数等扩增程序条件进行优化,寻找出适合此类菌物RAPD-PCR反应的最佳反应体系和最佳扩增程序。

Abstract:Genomic DNA of Hypoderma rubi was regarded as template optimized RAPD system of Hypoderma and its similar groups.The influencing factors and experimental parameters on RAPD were studied,including templates,dNTPs,Mg2+,primers,Taq DNA polymerase,annealing temperature and time,elongation time and cycle numbers.Optimal reaction system and amplification procedure were selected to apply to RAPD-PCR reaction of Hypoderma and its similar groups.
