Abstract:Copper-zine superoxide dismutase(SOD)from the erythrocytes of Chinese alligator has been purified by treatment with ethanol-chloroform mixture,K2HPO4 fractional salting-out, actone precipitation and DEAE-Sephadex A-50 column chromatography.Staining for SOD activity and by coomassie brilliant blue of the PAGE pattern of Purified SOD exhibits a single band.The molecular weight of purified SOD is about 32.8 kd as determined by gel filtration on Sephadex G-75 and its subunit molecular weight is about 16.2 kd as assayed with SDS-PAGE.It may be inferred that the enzyme is composed of two subunits of identical size.The enzyme subunit consists of about 153 amino acid residues,from which trypophan is absent.
