人单链白细胞介素12基因在果蝇细胞中的表达

摘    要:用PCR方法扩增不含信号肽序列的hscIL-12基因,并装载到果蝇表达质粒pMT/Bip/V5中,将其转化黑腹果蝇细胞S2中进行分泌表达,以淋巴细胞增殖实验检测表达的hscIL-12的生物学活性,结果表明hscIL-12可促进淋巴细胞增殖,呈剂量依赖性(P<0.05 ),提示在果蝇系统中表达的hscIL-12具有与天然IL-12相同的生物学活性,有良好的研究和临床应用前景。

Expression of Human Single Chain Interleukin 12 Gene in Drosophila Cells
Abstract:The hscIL 12 gene(no signal sequence), amplified by PCR, was subcloned into an expressing vector pMT/Bip/V5. The recombinant plasmid was transfered into Drosophila cell line S2. The secreted hscIL 12 fusion protein was identified by Western blot, and the bioactivity of hscIL 12 was investigated by lymphocyte proliferation assay. The results showed hscIL 12 expressed in S2 cells could stimulate lymphocyte proliferation in dose dependent manner. hscIL 12 has bioactivity identical to natural interleukin 12. These findings suggest that hscIL 12 is promising in theoretical reseach and in clinical use.
