Studies on the Structure-function Relationships of ANP Analogues

APIII rat atrial natriuretic peptide,ANP (1031-26)]and its analogues(A1-A7)were synthesized by the solid-phase method.The diuretic, natriuretic and hypotensive activities of these compounds were tested.Among these analogues,Al is 8.5 times as POtent as APIII in vasorelaxant activity and its diuretic and hypotensive activities were also more potent than APIII.A5 has only 1.5% of the potency of APIII in vasorelaxant activity.A2 and A7 have decreased potency in all three kinds of activities,but with their hypotensive activity(63%) less affected as compared with their diuretic activity(16%).The results indicate that the diuretic and hypotensive activities of ANP may be separated in two parts of the molecule and the activity of ANP is related to the whole structure of the molecule which is quite tight.
