摘    要:目的:研究" 促育生精方" 对精子特异性钙通道蛋白CatSper1、CatSper2 表达的影响。方法:采用Real-time PCR 法检测 CatSper1 mRNA、CatSper2 mRNA 在各组大鼠(模型组、低剂量组、中剂量组、高剂量组、空白对照组)精子中的表达;用Western blot 检测各组CatSper1 蛋白,CatSper2 蛋白的表达。结果:成功建立大鼠不育模型。CatSper1 mRNA、CatSper2 mRNA表达量为: 中、高剂量组显著高于模型组(P<0.05)。CatSper1、CatSper2蛋白表达量:中、高剂量组显著高于模型组(P<0.05)。结论:促育生精 方能有效提高少弱精症模型大鼠精子特异性钙通道CatSper1、CatSper2 基因及其蛋白的表达。

Effects of Cuyushengjingfang on the Expressions of CatSper1 and CatSper2 in Model Rats of Oligoasthenozoospermia Induced by Cyclophosphamide
Abstract:Objective:To investigate the effect of "Cuyushengjingfang" on the expression of CatSper1 and CatSper2 in the sperm ofOligoasthenozoospermia Wistar rats induced by Cyclophosphamide (CP).Methods:Thirty male Wistar rats were randomly divided into model group(MG), low-dose Cuyushengjing group(LDCG), middle-dose Cuyushengjing group(MDCG), high-dose Cuyushengjing group (HDCG) and blank control group(BCG). Model rats of oligoasthenozoospermia were created by CP (30 mg/kg) injected into the abdominal cavity of rats once a day for 5days, while BCGwere injected with normal saline (NS). The rats in LDCG,MDCGand HDCGwere oral administrated with 3ml Cuyushengjingfang for 35 days while the rats in MG and BCG were oral administrated with 3ml NS. The concentration of Cuyushengjingfang in that of LDCG, MDCG and HDCG were 0.84 g/ml, 1.69 g/ml, and 3.37 g/ml. The proteins expression of CatSper1 and CatSper2 in sperm was measured byWestern blotting and Real-time PCR.Results:Models rats of Oligoasthenozoospermia were created successfully. The mRNA expression of CatSper1 and CatSper2 in MDCG and HDCG were significantly higher than that in MG (P<0.05). The protein expression of CatSper1 and CatSper2 in MDCG and HDCG were higher than that in MG (P<0.05).Conclusion:Cuyushengjingfang could effectively increase the expression of CatSper1 and CatSper2 in model Rats of Oligoasthenozoospermia created by Cyclophosphamide.
