辣椒碱对3T3-L1 前脂肪细胞葡萄糖摄取的影响

摘    要:目的:探讨辣椒碱对3T3-L1前脂肪细胞葡萄糖摄取的影响。方法:不同浓度的辣椒碱作用于3T3-L1前脂肪细胞,采用MTT测定细胞活性,GLU Test试剂盒法测定葡萄糖摄取,Western Blot法检测葡萄糖转运蛋白1(GLUT-1)表达的变化。结果:25μM辣椒碱作用72 h和50μM、100μM辣椒碱作用48 h、72 h,可显著抑制3T3-L1细胞增殖,6.25、12.5、25μM辣椒碱作用可显著促进3T3-L1细胞的葡萄糖摄入,Western Blot结果显示辣椒碱能够显著增加GLUT1蛋白表达量,差异均具有统计学意义(P0.05)。结论:低剂量辣椒碱具有降糖作用,其作用机制可能与增加GLUT-1蛋白表达有关。

Effects of Capsaicin on Glucose Uptake in 3T3-L1 Preadipocytes
Abstract:Objective:To evaluate the effect andpotential mechanism of capsaicin on the glucose uptake in 3T3-L1 preadipocytes in vitro.Methods:3T3-L1 preadipocytes cells were exposed to different concentrations capsaicin for 24 h, 48 h and 72 h. MTT assay was used to detect their effect on the cell vitality. Glucose metabolism was determined by GLU Test Kits. The GLUT-1 protein was demonstrated by Western Blot.Results:Treatment of capsaicin (25 uM, 50 uM, 100 uM) showed significant effect on cell viability. Capsaicin (6.25, 12.5, 25 uM) could improve glucose uptake in 3T3-L1 cells. Western Blot results showed that the glucose transporter 1 (GLUT-1) protein expressions were significantly increased in treatment groups than control.Conclusion:the effect of capsaicin on glucose metabolismmight be associated with promoting the GLUT-1 protein expression.
