摘    要:目的:探讨转染Canstatin基因对人肝癌HepG-2细胞体外形成血管生成拟态的影响。方法:将Canstatin基因通过脂质体法转染人肝癌HepG-2细胞,行G418筛选获得转基因细胞克隆。用SDS-PAGE电泳检测Canstatin蛋白在转基因细胞培养上清液中的表达;建立HepG-2细胞人工基底膜基质凝胶体外三维细胞培养模型,观察HepG-2细胞能否形成血管生成拟态,并比较转基因和未转基因细胞的管道形成能力。结果:Canstatin在转染人HepG-2细胞中表达并分泌至上清液中,人肝癌HepG-2细胞在体外三维培养条件下能够形成血管生成拟态。Canstatin基因转染HepG-2细胞组的管状结构数量高于空载体组和HepG-2细胞组,转染细胞管道形成能力明显受抑制。结论:人肝癌HepG-2细胞株可形成血管生成拟态,Canstatin能抑制人肝癌细胞株HepG-2体外血管生成拟态形成。

Abstract:Objective: To investigate the effect of Canstatin on vasculogenic mimicry in human hepatocellular carcinoma cell line HepG-2 in vitro. Methods: Expression vector of pcDNA3.1-Canstatin-3FLAG was transfected into HepG-2 cells by liposome, and the positive clone was screened by G418.
