散发性阿尔茨海默病大鼠学习记忆和抗氧化能力的观察

摘    要:目的:观察散发性阿尔茨海默病动物模型的学习记忆和抗氧化能力,初步探讨该疾病的发生机制。方法:雄性SD大鼠24只,随机分为正常对照组和模型组。模型组大鼠给予侧脑室注射STZ制作散发性阿尔茨海默病动物模型,3周后,通过Morris水迷宫方法进行行为学检测,生物化学方法检测海马超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量的变化。结果:与对照组比较,模型组大鼠潜逃潜伏期延长(P<0.05)、目的象限停留时间显著降低(P<0.05);海马SOD活力降低(P<0.01),MDA含量增加(P<0.01)。结论:散发性阿尔茨海默病大鼠学习记忆能力低,其发生机制可能与大鼠海马的抗氧化能力下降有关。

Study of Learning,Memory and Antioxidant Capacity of Hippocampus in Rat Models of Sporadic Alzheimer's Disease

Abstract:Objective:To establish experimental sporadic Alzheimer's disease rat model and to investigate the effects of intracere- broventricular (ICV) administration of streptozotocin (STZ) on SOD and MDA in rat hippocampus.Methods:Twenty-four SD rats were randomly divided into control group and model group.The rats in the model group received intracerebroventricular injection of STZ bi- laterally with a dose of 3 mg/kg and the injection was repeated after 3 days,and the rats in control group received the same volume of ar- tificial cerebrospinal fluid as the model group.Three weeks after the first injection of STZ or substitute,the animals were tested by Morris water mass to observe the function of learning and memory.Then the rats were killed for examining the changes of anti-oxidative ability in hippocampus.Results:Compared with the control group,the rats in model group represented more escape latency (P<0.05) and less time spent in the right quadrant (P<0.05).At the same time,the SOD activity decreased and MDA increased in rat hippocampus in model group (P<0.01).Conclusions:The function of learning and memory in rats with Alzheimer's disease decreased.Its machanism is possibly related to the dysfunction of anti-oxidative ability of hippocampus.
