TGF-beta1 基因rs200482214 位点多态性与黑龙江省汉族人群慢性牙周炎的相关性研究

目的:探讨转移生长因子β1(transforminggrowth factor beta-l,TGF-β1)位点rs200482214基因多态性与黑龙江省汉族人群慢性牙周炎的易感性的相关性。方法:选取2012年3月至2013年7月在哈尔滨医科大学附属口腔医院牙周科就诊的135例轻、中、重度慢性牙周炎汉族患者(牙周炎组)和108例汉族健康对照者(健康对照组)作为研究对象,基因组DNA来自口腔颊粘膜拭子,采用多重单碱基延伸SNP分型技术(Multiplex SNaPshot technique)对所有受试者TGF-β1基因rs200482214位点进行检测,比较两组间此位点基因型分布和等位基因频率的差异。结果:(1)TGF-β1基因rs200482214位点各基因型(GG、GA、AA)分布均符合Hardy-Weinberg遗传平衡定律(P>0.05);(2)TGF-β1基因rs200482214位点GG、GA、AA在牙周炎组和健康对照组的分布频率分别为61.5%、30.4%、8.1%和63.0%、28.7%、8.3%,两组人群基因型分布频率差异无统计学意义(P>0.05);等位基因G、A在牙周炎组和健康对照组分布频率分别为76.7%、23.3%和77.3%、22.7%,两组人群的等位基因分布频率差异亦无统计学意义(P>0.05)。结论:TGF-β1位点rs200482214基因多态性与黑龙江省汉族人群慢性牙周炎的易感性不具有相关性。

Association between TGF-beta1 rs200482214 Polymorphism and Susceptibility to Chronic Periodontitis in Chinese Han in Heilongjiang

Objective:To investigate the association between TGF-beta1 rs200482214 polymorphism and susceptibility to Chronic periodontitis in Chinese Han in Heilongjiang.Methods:We performed a case-control study on 135 mild, moderate, severe chronic periodontitis patients and 108 healthy controls. Genomic DNA from oral mucosa of two groups was got. Polymorphism of TGF-beta1 gene was analyzed using the Multiplex SNaPshot technique, the differences of the genotype and allele frequencies were calculated and analyzed.Results:(1)All genotypes distribution of the TGF-beta1 rs200482214 polymorphismwere in Hardy-Weinberg equilibrium in both the cases and the controls; (2)TGF-beta1 rs200482214 GG, GA, and AA genotype frequencies were 61.5 %, 30.4 % and 8.1 % in case samples, and 63.0 %, 28.7 % and 8.3 % in control samples, respectively, two groups genotype frequencies distribution were not significantly different (P>0.05); The allele G, A distribution frequencies were 76.7 %, 23.3%in the periodontitis group and 77.3 %, 22.7 % in the control group, no significant difference was found in the distribution of allele frequencies between two groups (P>0.05).Conclusion:TGF-beta1 rs200482214 polymorphism may be not association with the susceptibility to Chronic Periodontitis in Chinese Han in Heilongjiang.
