摘    要:从系统进化树重建原理出发,比较了HCV常用的几个基因分型区域的分型效果,包括5′UTR、core、E1、E2和NS5B区,探讨最能代替全基因组基因分型的区域.结果发现以5′UTR区建树,基因分型不完全正确而以core区、E1区、E2区及NS5B区建树,基因分型均完全正确,但同一基因型间的核苷酸演化距离存在差异.计算5条1a型序列的core区、E1区、E2区、NS5B区的核苷酸演化距离并和全基因组序列核苷酸演化距离比较,结果发现NS5B蛋白区基因分型最能反映病毒株的演化关系.确定NS5B区为丙型肝炎病毒基因分型的最佳区域.

Abstract:Prom the phylogenetic tree reconstruction principle,we compared the HCV genotyping of several commonly used in the sub-region type effects,including the 5'UTR,core, E1,E2,and NS5B areas,to explore the best instead of whole-genome genotyping of regional.The results are found to have 5'UTR District achievements,genotyping is not entirely correct to core areas,E1 area,E2 area and NS5B District achievements,genotyping are completely correct,but the same genotype evolution of the distance between the nucleotide differences in.Calculation 5 1a-type sequence of core areas,E1 area,E2 area,NS5B region and the nucleotide distance and evolution of whole-genome nucleotide sequence of evolutionary distance comparison and found that NS5B protein gene typing of best reflects the strain of the virus evolutionary relationships. Determine the area of hepatitis C virus NS5B genotyping of the best area.
