C型肉毒毒素的制备及类毒素保护力初探

摘    要:将C型肉毒梭菌经适宜条件的产毒培养后纯化,并进行相关鉴定。制备的C型肉毒毒素用分段脱毒法脱毒,并进行类毒素保护力的初步研究。以不同蛋白含量C型肉毒类毒素免疫小鼠后攻毒,结果显示,蛋白含量为0.625μg的类毒素免疫2针或蛋白含量为1.25μg的类毒素免疫1针均可保护50LD50的C型肉毒毒素攻击。蛋白含量为5μg的C型肉毒类毒素与福氏不完全佐剂配制的抗原免疫小鼠3次所得抗血清的保护力(Anti LD50/ml)为4.3×104。说明用该纯化工艺制备的C型肉毒类毒素具有很好的免疫原性,作为抗原成分用于C型肉毒疫苗和C型肉毒抗毒素的研究和生产具有较好的应用潜力。

Preparation and characterization of Clostridium botulinum type C and primary evaluation of its toxoid's protective effects
Abstract:Preparation toxin of Clostridium botulinum type C from strain 6514,for which are practical importance in further development of multi-application. Identification was carried out after the Clostridium botulinum type C were cultured and purified. After the purified toxin inactivated with formaldehyde the experiments were followed: One group used Al(OH)3 as the adjuvant and toxoid in different amount. Mice were immunized with different doses. After immunization the mice were challenged with 50LD50 toxin of BONT/ C.The other group was used FIA as the adjuvant in detection of the value related to the antiserum. Challenge test indicated that the toxoid group of 0.625μg immuned twice or 1.25μg immuned once can protect animals from the challenge of 50LD50 of BONT/ C. The average value of the antiserum obtained from immunized mice three times with toxoid group of 5μg was about 4.3×104LD50.The purified toxin of Clostridium botulinum type C has a good immunity,suitable to be used as the antigen in vaccine preparation or antitoxin production.
