芍药花瓣总RNA的提取

摘    要:用TRIzol法和改良的热硼酸盐法从芍药幼嫩花瓣中提取总RNA,总RNA的D260nm/D280nm值分别为1.803、1.974。琼脂糖电泳表明,用TRIzol法提取的RNA只有28S、18S2条带,带的亮度差,说明RNA有些降解;而用改良的热硼酸盐法提取的总RNA有28S、18S、5S3条带,带的亮度强,且28S是18S宽度的2倍左右,说明提取的RNA完整、未降解,可用于后续实验。改良的热硼酸盐法更适于从芍药花瓣中提取总RNA。

Abstract:Total RNA from the tender petals of Paeonia lactiflora Pall was extracted by the methods of TRIzol and improved hot-borate. The results showed that the D260nm/D280nm of two methods were 1.803 and 1.974 respectively. After gelose electrophoresis, the RNA extracted by TRIzol only had two bands 28S and 18S, and the bands were dark, which meaned RNA is destroyed more or less; while the RNA extracted by improved hot-borate had three bands 28S, 18S and 5S, the bands were light, and the 28S was as two times length as the 18S, which meaned the extracted RNA was integrated enough to be used into the following experiment. The method of improved hot-borate is more suitable to extract total RNA from the tender petals of Paeonia lactiflora Pall.
