宁夏枸杞DFR基因的克隆与序列分析

摘    要:根据已报道植物二氢黄酮醇4-还原酶(dihydroflavonol 4-reductase,DFR)基因cDNA序列的保守区域设计引物,对宁夏枸杞(Lycium barbarum)DFR基因进行克隆及序列分析.结果表明,宁夏枸杞DFR基因cDNA全长1140个碱基,有一个1116个核苷酸的开放阅读框,编码一个长372个氨基酸的蛋白质;氨基酸序列的聚类分析显示,宁夏枸杞LbDFR1与马铃薯、烟草、蕃茄等茄科植物的DFR亲缘关系较近,其中与马铃薯DFR亲缘关系最近,相似性为92%;通过编码蛋白的三级结构预测,该蛋白为单体模式,具有酶学的生物特征;宁夏枸杞LbDFR1在宁夏枸杞的根、茎、叶等组织中广泛表达,为组成表达型基因.

Abstract:Degenerate primers designed according the conservative domain of dihydroflavonol 4-reductase(DFR) in plants were used to amplify the DFR fragments from cDNA of Lycium barbarum.The full-length cDNA of DFR from L.barbarum had 1 140 bp with an open reading frame(1~1 116 bp),encoding 372 amino acids of protein.Hylogenic analysis on the amino acid sequence of DFR from L.barbarum with those of other plants showed that L.barbarum was closely related to Solanum tuberosum,S.pinnatisectum,S.lycopersicum,and Nicotiana tabacum,espically S.tuberosum,which similarity related to 92%.The protein was monomer model.LbDFR1 was constituted expressed gene in root,stem,and leaf of L.barbarum.
