| 摘 要： | Dasabuvir 是 2015 年 1 月 15 日以单药成份获得欧洲药品管理局批准的抗丙肝新药，该药于 2014 年 12 月 19 日以复方药形式
（dasabuvir/ombitasvir/paritaprevir/ritonavir）获得美国食品药品管理局批准用于治疗基因 1 型慢性丙肝病毒感染，包括有代偿性肝
硬化患者的治疗。Dasabuvir 是一种 HCV RNA 聚合酶 NS5B 抑制剂，干扰 HCV 的正常复制而发挥抗病毒作用。介绍 dasabuvir 的化学
合成、临床前药理学研究、临床研究及专利保护情况等，为抗丙肝新药的研发提供参考。

| Abstract: | On Jan 15, 2015, dasabuvir was approved by European Medicines Agency as monotherapy for the treatment of HCV. The drug was also
approved by FDA on Dec 19, 2014 as combined therapy (dasabuvir/ombitasvir/paritaprevir/ritonavir) for the treatment of HCV genotype 1. Dasabuvir
is a NS5B RNA-dependent RNA polymerase inhibitor, which exerts anti-viral activity by interfering with HCV replication. The chemical synthesis,
preclinical pharmacology, clinical trials and patent protection of dasabuvir were summarized in this article, so as to provide references for the R&D of
new anti-HCV drugs. |
