| 摘 要： | 本文描述了一种新的、简化了的纯化T_4RNA连接酶的方法——免疫亲和层析法.利用这种纯化方法得到的酶,经SDS-凝胶电泳,得到一条主带。将此酶与~(32)pCp及CpGpGpA(或tRNA)一起进行连接反应,未见到RNase杂酶的降解作用。用此方法制备的酶,可用于确定顺序的核酸的合成。

| Abstract: | A simplified purification method of the T4 RNA ligase using immunoaffinity chromatography is described. The purified T4 RNA ligase produced one major band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. 32pCp and CpGpGpA (or tRNA) were incubated with the purified enzyme and subjected to homochrom atography.No degradation products were found, and this purified enzyme could be used in the synthesis of nucleic acids with defined sequence. |
