Abstract:
A digoxigenin (DIG)-labelled rRNA probe was used to hybridize in situ to the vibration sections of paraformaldehyde fixed root tips, followed by immunogold staning and silver enhancement treatment After the vibration sections which had undergone in situ hybridization, immunogold staining and silver enhancement were observed and photographed under the light microscope, they were further prepared into ultrathin sections and studied underthe electron-microscope. This method more directly combined the in situ hibridization at level of the light microscope with that at level of the electron microscope, by using a piece of vibration section, through which a more detailed rDNA localization was revealed in interphase nuclei of Pisum sativum. Some advantage and disadvantage about the method is discussed in this paper.
