从龙眼鬼帚病树提纯一种线状病毒

PARTIAL PURIFICATION OF A FILAMENTOUS VIRUS FROM LONGAN ( DIMOCARPUS LONGANA LAM. ) WITCHES' BROOM DISEASE TREES

It is the first report on the purification of a filamentous virus from longan trees with witches' broom disease. Deep freezed leaves or barks were pulverized in a mortor, and Tris buffer added. After homogenation, the sap was squeezed through cheesecloth, clarified by chloroform, concentrated by two cycles of PEG precipitation and differential centrifugation, and fractionated by sucrose density gradient centrifugation. The position of virus layer was detected and located by a UV monitor at 254nm. The partially purified virus was stained by 2% UA and examined under electron microscope. It contains a number of the filamentous virions with a diameter of about 15nm and a lenth of 300-2,500nm. The main distribution of the virus particles is 700-l,300nm. No virus Particle was detected in healthy plants. Rabbit antiserum against the virus was prepared and used to trap the virions from the infected longan leaves by SPA-ISEM technique.
