摘    要:通过海藻酸钠/纤维素硫酸钠-聚二甲基二烯丙基氯化铵(SA/NaCS-PDMDAAC)微胶囊固定化酵母细胞将胞苷一磷酸(CMP)转化为胞苷三磷酸(CTP),考察了各种因素条件对CTP转化率的影响,以提高CTP的转化率.通过考察分批补料添加葡萄糖,固定化酵母量,CMP浓度等以达到提高CTP转化率的要求.结果在250 mL锥...

Abstract:Cytidine monophosphate(CMP) was converted to cytidine triphosphate by SA(sodium alginate) / NaCS(cellulose sulphate)-PDMDAAC(poly dimethyl diallyl ammonium chloride) microcapsule immobilized yeast cells.To observe effect of all factors on the synthesis of CTP and raise the conversion rate of CTP,the fed-batch glucose,the amount of immobilized yeast and CMP concentration were examined.In this experiment 250 mL-Erlenmeyer flask with 20 mL reaction liquid including 60 mmol·L-1 CMP,150 mmol·L-1 glucose,250 mmol·L-1 phosphate buffer,20 mmol·L-1 magnesium chloride,pH6.5 was used.25g immobilized yeast was added and incubated at 35℃.Then added 0.27g glucose per reaction of 2h.When total reaction time reached 6h,about 80% CMP was transformed to CTP.The immobilized yeast cells could be repeatedly used for 5 times,and the conversion rate remained stable at around 70%.
