Modification of hepatic microsomal oxidative drug metabolism in rats by the opiate maintenance drugs acetylmethadol, propoxyphene, and methadone.

Abstract:The formation of cytochrome P-450 “metabolic intermediate” complexes invivo occurred with acetylmethadol and propoxyphene, but not methadone in both naive and phenobarbital-induced animals. The invivo formation correlated with the relative ability of these three compounds to form metabolic intermediate complexes and inhibit mixed-function oxidation reactions in vitro.
