一株絮凝菌的鉴定、絮凝基因定位及其絮凝剂成分分析

摘    要:从武汉市综合废水处理的活性污泥中筛选得到一株絮凝剂产生菌株MBF03,其絮凝率达到90%以上,絮凝效果稳定.通过16S rDNA鉴定,该菌为泛菌属,扫描电镜结果显示为杆状.通过质粒转化与质粒消除试验,初步证实了MBF03菌的絮凝基因位于染色体上.提取该菌所产的絮凝剂,进行紫外、红外分析,结果表明,该絮凝剂主要成分是胞外多糖类物质,不含蛋白质和核酸.

Abstract:One flocculant-producing strain named MBF03 was isolated from the activated sludge of Wuhan integrated wastewater treatment factory,of which flocculation rate reached 90%.Sequencing of the 16S rDNA gene revealed identification as Pantoea sp..Scanning electron microscopy showed that MBF03 was rod-shape.With plasmid transformation and plasmid elimination experiments,we primary confirmed that the flocculation gene of MBF03 bacteria was located on chromosome DNA rather than a plasmid.Subsequent ultraviolet and infrared analysis on MBF03 flocculants revealed the main component of the extracellular flocculants was polysaccharide rather than protein and/or nucleic acid.
