摘    要:
Burkitt淋巴瘤和未分化型鼻咽癌的细胞中存在着Epstein-Barr病毒(EBV)的基因组,在前者虽经长期体外培养,病毒基因组仍持续存在,并部分地表达。这些现象提示,至少有一些EBV基因组是整合在寄主细胞的DNA中。Kieff等用染色体原位杂交检测Burkitt淋巴瘤Namalwa细胞株和EBV转化的人脐带血B淋巴细胞IB_4株,发现病毒DNA是整合在前者的1号染色体上,在后者则整合

Abstract:
Using[32P]-dCTP.[32P]dTTP labeled EBVDNA BamH I -W fragments cloned in pBR322as probe,blot hybridization was carriedout with DNA extracted from forty nasopha-ryngeal biopsies (31 cases of nasopharyngeal 6carcinoma,cases of nasopharyngitis,3 sam-ples of nasopharyx mucosa from inducedabortive fetus) obtained in NPC endemicarea,South China.The results showed that,in the NPC group,about 87% was viralDNA positive,while there were three biop-sies,diagnosed as nasopharyngitis,also con-taining EBV DNA.In accordance with ourprevious experiments,assaying for EBNA byACIF method,the two NPC epithelioid celllines CNE,and CNE_2,were hybridizationnegative (Table I ). Autoradiograms of DNAs isolated fromhybridization positive NPC biopsies after di-gestion with EcoR I showed that all slotspresented only one band corresponding toEcoR I A fragment,except one with an ad-ditional band having a molecular weight lar-ger than A (Fig.2.slot 18).Since thereare no EBV DNA EcoR I fragments otherthan A having homologous sequence withBamH I W,the presence of this novel bandmay result from integration of viral DNAinto host cell genome,leading to the appea-rance of a new EcoR 1 cleavage site.
