摘    要:通过单因素实验和正交实验对影响桦褐孔菌三萜提取的异丙醇体积浓度、硫酸铵质量浓度、料液比和超声时间等因素进行探讨,旨在建立桦褐孔菌Inonotus obliquus菌丝体三萜类化合物的提取工艺。结果表明,提取桦褐孔菌三萜的最佳条件为:异丙醇体积浓度40%,硫酸铵质量浓度0.125g/mL,料液比(W/V)0.9:1,超声时间为35min,三萜提取率为6.18%,提取效率明显高于常规超声波提取法(5.38%)。利用该方法获得的三萜具有较强清除自由基的能力。通过正交实验获得的桦褐孔菌三萜的提取工艺是一种经济、有效的方法。

Extraction of triterpenoids from Inonotus obliquus by aqueous two-phase systems associated with ultrasonic extraction

Abstract:This study was designed to optimize extraction of triterpenoids from the mycelia of Inonotus obliquus by orthogonal test method. Extraction of triterpenoids was probed by single factor experiment and orthogonal design experiment. The range of the independent variables, namely levels of isopropyl alcohol and ammonium sulfate, ultrasonic duration and material-solvent ratio were identified by a first set of single factor experiments. The actual values of the independent variables coded at three levels were selected based on the results of single factor experiments. Subsequently, the levels of isopropyl alcohol and ammonium sulfate, ultrasonic time and material-solvent ratio were optimized using the orthogonal array method. The optimum conditions for the extraction of triterpenoids were as follows, isopropyl alcohol concentration 40%, the amount of ammonium sulfate 0.125g/mL, material-solvent ratio 0.9:1 (W/V), and ultrasonic duration 35min. The extraction rate of triterpenoids was 6.18% in the optimum extraction conditions, much higher than that of ultrasonic extraction (5.38%). The triterpenoids compounds obtained by this method had stronger antioxidant activities than those by ultrasonic extraction method. This extraction method is an economic and effective process for extracting triterpenoids from the mycelia of I. obliquus.
