摘    要:研究了类卟啉稀土配合物(以下简称PLM-Gd-A)对小鼠腹水肝癌细胞(AH)的光敏作用及AH细胞对PLM-Gd-A的摄取表明:PLM-Gd-A被AH细胞摄取的速度快,用MTT方法测定了细胞光敏存活曲线,杀伤细胞的能力与光照时间和光照强度以及PLM-Gd的浓度密切相关,用FADU方法和电镜观察结果证明:该光敏损伤细胞的靶主要是在细胞核;对于不同稀土离子配合物光敏能力比较表明:Gd>Eu>Sm;造成细

PHOTOSENSITIZING DAMAGE OF PORPHYRIN-LIKE EARTH COMPLEX(ES) (PLM-Gd) TO MURINE ASCITIC HEPATOMA CELLS
Abstract:The porphyrin-like macromolecules and their rare earth metal complexes (PLM-Complex) belong to a noval family of dye molecules, they absorb strongly around 760-780nm, which were first suggested as potential photosensitizers for Photo Dynamic therapy (PDT) by J.L. Sessler in 1987. Up to now their photo sensitizing damage to cells have not been studied. In this paper we studied the PLM-Gd photosensitized cytotoxity to Murine ascitic hepatoma cell(AH). The results showed that the cellular uptake of PLM -Gd is a complicated process. The saturation uptake concentration is aboat 1× 109 molecular/each each cell of AH cells. The cell mortality was proportional to PLM-Gd concentration upon illumination at 760 nm wavelength. The ability to kill AH cell among the PLM-Gd;PLM-Eu; PLM-Sm and PLM-Gd-B are in the following order: PLM-Gd-B;PLM -Gd-A; PLM- Eu; PLM -Sm. With Fluorometric analysis of DNA unwinding method for detection of DNA strand breaks in cell, the results showed that DNA strand breaks remarkably. By using 1O2 quenchers(L-His,NaN3), superoxide anion radicals quencher (SOD), we proved that singlet oxygen and surperoxide anion were involved in the process of PLM-Gd photosensitized cytotoxicity .
