LXRs激动剂T0901317对成人骨骼肌细胞FAT/CD36基因mRNA表达的影响

摘    要:目的:探讨肝核受体LXRs激动剂T0901317对正常人骨骼肌细胞中FAT/CD36基因mRNA表达的影响。方法:将原代培养的5例成人骨骼肌细胞分为用肝核受体LXRs激动剂39901317(1μmol/L)作用组、T0901317(0.5μmol/L)作用组和阴性对照组,作用24h后采用sYBR Green Ⅰ实时荧光定量PCR法检测各组成人骨骼肌细胞FAT/CD36基因mRNA表达水平,并用2^-^△△Ct方法进行比较分析。结果:①以浓度为1μmol/L的T0901317作用组和0.5μmol/L的T090131作用组和对照组样本的均数进行方差分析,差别有统计学意义(P〈0.01)。②浓度为1μmol/L的T0901317作用组和0.5μmol/L的T090131作用组成人骨骼肌细胞中FAT/CD36基因的mRNA表达分别是对照组的3.03倍和2.91倍。结论:肝核受体LXRs激动剂T0901317能够提高成人骨骼肌细胞中FAT/CD36基因mRNA的表达水平,提示30901317有加快骨骼肌细胞内脂肪酸的堆积作用,推测IXRs激动剂T0901317可能会增加糖尿病患者骨骼肌胰岛素抵抗的风险。

Abstract:Aim: To investigate the liver X receptors agonists T0901317' s effect on expression of FAT/CD36 gene mRNA in adult human skeletal muscle cell. Methods: Myotubes from humans were exposed to different T0901317 concentrations(0, 0.5, and 1.0μmol/L) for 24 hours before experiments were performed. Then the expression of FAT/CD36 mRNA in skeletal muscle cell of each experimental group was detected by SYBR Green Ⅰ real-time quantitative polymerase chain reaction. The relative data were compared among groups by 2^-△△Ct method. Results: ①The Ct mean of control group,T0901317(0.5 μmol/L) group, T0901317(1 μmol/L) group were analyzed and there was significant differenee(P 〈 0.01).②The expression of FAT/CD36 mRNA with liver X receptors agonists T0901317 in human skeletal muscle cell in the 39901317(0.5/.anol/L) group and T0901317(1/nnol/L) group were 2.91 times and 3.03 times than the control group. Conclusion: The expression of FAT/CD36 mRNA in human skeletal muscle cell afer the treatment of fiver X receptors agonists T0901317 is increased, so we may propose that T0901317 may increase the risk of resistance in adult human skeletal muscle.
