摘    要:目的研究人乳头状瘤病毒(HPV16)在乳腺癌中的表达及作用机制,检测HPV16、诱导一氧化氮合酶(iNOS)及P53蛋白在乳腺癌的表达及其间的相关性。方法应用免疫组化SP法,共检测了52例乳腺癌和16例乳腺良性瘤HPV16、iNOS和P53蛋白的表达。结果HPV16和P53蛋白在乳腺癌组的阳性表达率均显著高于乳腺良性瘤组。统计分析表明,iNOS和P53蛋白表达阳性率与HPV感染率密切相关(P〈0.05)。结论HPV16感染参与了乳腺癌的发生,其致癌机制可能是通过诱导iNOS表达增加,产生NO,诱导p53基因突变,从而导致乳腺癌的发生。

Abstract:Objective To explore the expression of human papillomavirus (HPV) in mammary carcinoma and its function in carcinogenesis,the expression of HPV-16, iNOS and P53 were investigated. Method 52 samples of mammary carcinoma and sixteen samples of benign breast tumor were assayed using immunohistochemical SP method for detection of HPV-16,iNOS and P53 protein expression level. Results The expression of HPV-16,iNOS and P53 of mammary carcinoma were significantly higher than the benign group. Analysis showed that the expression of iNOS and P53 were significantly correlated with the infection of HPV16 in mammary carcinoma ( P 〈 0.05). Conclusions HPV might play an important role in the process of carcinogenesis of mammary carcinoma in Harbin. Its mechanism may be as follows : the infection of HPV may promote the expression of iNOS. NO produced by iNOS may induce the mutation of wild type p53. This may promote the mammary carcinogenesis.
