转柽柳eIF1A基因烟草的耐盐性分析

摘    要:
目的:验证柽柳eIF1A基因的功能,为通过基因工程手段培育耐盐植物提供基础资料。方法:对转eIF1A基因的烟草和对照烟草进行不同浓度NaCl胁迫实验,测定其相对电导率、SOD活性和丙二醛含量,统计生根率、生长量和盐害程度。结果:转基因烟草的相对电导率、丙二醛含量均随盐浓度的增加而增大,但都较非转基因对照烟草低。SOD活性随着盐浓度的升高而升高,相同浓度NaCl胁迫下各转基因烟草的SOD活性均高于对照烟草的SOD活性。NaCl浓度为240mmol/L时,非转基因对照烟草不能生根,盐害指数高达67.7%;而转基因烟草均能生根,大部分转基因株系的生根率大于50%。结论:柽柳eIF1A基因的转化提高了烟草的耐盐性。

Abstract:
Objective:To analysis the functions of eIF1A gene from Tamarix sp.in order to cultivated salt tolerance plants through the genetic engineering means.Study on the effects of different NaCl concentration stress to the tissue culture of transgenic tobaccos with eIF1A gene from Tamarix sp.Methods:The relative electrical conductivity,SOD activity,MDA content,rooting efficiency,growth capacity and salt injury degree of the transgenic tobaccos and control were measured.Results:The results showed that the relative electrical conductivity and MDA content of the transgenic tobaccos were increased with salt solution,but lower than control;the SOD activity of the transgenic tobaccos was increased with salt solution,but higher than control.Under 240 mmol/L NaCl concentration,the transgenic tobacco rooted well,rooting efficiency more than 50%;but the control could not produce roots and the salt injury was 67.7%.Conclusion:All the results proved that the eIF1A gene from Tamarix sp.could enhance the salt tolerance of transgenic tobacco.
