Two polymorphic dinucleotide repeats in rainbow trout (Oncorhynchus mykiss)

Abstract:Source/description: A genomic plasmid library was constructed in the plasmid vector pCR-ScriptTM (Stratagene, La Jolla, CA) from size-selected (300–800 bp) rainbow trout genomic DNA digested with PvuII and HaeIII. The library was screened with a (TG)9 probe. Positive clones were sequenced using an automatic DNA sequencer (A.L.F. express DNA Sequencer, Pharmacia, Uppsala, Sweden). The clone Omy0004DIAS was shown to contain a (GT)17 repeat (GenBank accession number AF090389 ), while clone Omy0008DIAS contained a (GT)11 repeat (GenBank accession number AF090390 ). PCR primers were designed on the basis of the flanking sequences. The sizes of the PCR fragments were analysed on the A.L.F. express DNA Sequencer.
